Enrico Luchinat è nato nel 1985. Si è laureato nel 2007 in Chimica cum laude nel 2009 e in Chimica delle Molecole Biologiche cum laude presso l'Università di Firenze. Nel 2013 ha conseguito il dottorato di ricerca “International Doctorate in Mechanistic and Structural Systems Biology” presso la stessa università, sotto la supervisione della Prof.ssa Lucia Banci. È stato borsista post-dottorato (2013-2019) presso il Dipartimento di Scienze Biochemiche Sperimentali e Cliniche e ricercatore presso il Dipartimento NEUROFARBA (2020-2021). Nel 2022 è entrato a far parte del Dipartimento di Scienze e Tecnologie Agro-alimentari dell'Università di Bologna come RTDa, e nel 2024 è entrato nel Dipartimento di Chimica dell'Università di Firenze come RTDb.
È co-autore più di 45 pubblicazioni peer-reviewed ed è stato invitato come relatore in oltre 30 conferenze internazionali e eventi virtuali. Nel 2020 ha ricevuto il GIDRM Under-35 Award per lo sviluppo di approcci innovativi di NMR in cellula.

La sua ricerca si concentra sullo sviluppo e sull'applicazione di metodi di spettroscopia di risonanza magnetica nucleare in cellula (in-cell NMR) per studiare la struttura, la dinamica e le interazioni delle proteine a livello atomico all'interno di cellule vive. Ha contribuito a sviluppare un approccio innovativo basato sull'espressione di proteine marcate isotopicamente in cellule umane, che consente di studiare i processi di folding e maturazione delle proteine a livello atomico. Ha inoltre contribuito allo sviluppo di un bioreattore NMR in flusso in grado di preservare la vitalità delle cellule per diversi giorni, e alla sua applicazione allo studio di eventi intracellulari in tempo reale.
Attualmente si concentra sullo studio delle interazioni proteina-ligando all’interno della cellula, utilizzando approcci in-cell NMR sia per osservare le proteine che i ligandi. Sta inoltre sviluppando nuovi schemi di marcatura isotopica per le cellule umane, basati sull'incorporazione di amminoacidi fluorurati e sull'uso di precursori marcati di specifici amminoacidi.
La combinazione di queste metodologie consente di caratterizzare aspetti strutturali e meccanicistici di macromolecole biologiche nel loro ambiente cellulare, con grande rilevanza fisio-patologica.

Enrico Luchinat was born in 1985. He graduated in 2007 with a B.Sc. in Chemistry cum laude and in 2009 with a M.Sc. in Chemistry of Biological Molecules cum laude at the University of Florence. In 2013, he obtained a Ph.D. in the International Doctorate of Mechanistic and Structural Systems Biology at the same University, under the supervision of Prof. Lucia Banci. He has been Postdoctoral fellow (2013-2019) at the Department of Experimental and Clinical Biochemical Sciences and Research fellow at NEUROFARBA Department (2020-2021). In 2022, he joined the Department of Agricultural and Food Sciences and Technologies at the Bologna University as RTDa Junior Assistant Professor, and in 2024 he joined the Chemistry Department of the University of Florence as Tenure-track Assistant Professor (RTDb).
He has co-authored more than 45 peer-reviewed publications and has been invited as speaker at more than 30 international conferences and virtual events. In 2020, he was awarded the GIDRM Under-35 Award for the development of innovative in-cell NMR approaches.

His research focuses on the development and application of in-cell NMR spectroscopic tools to study protein structure, dynamics and interactions at the atomic level inside intact, living cells. He contributed to pioneer an approach based on the direct expression of isotope-labeled proteins in cultured human cells, which allows studying protein folding and maturation processes at the residue-level. He contributed to develop a modular flow NMR bioreactor capable of preserving cell viability for several days, and applied it to monitor intracellular events in real time.
He is currently focusing on the study of intracellular protein-ligand interactions, both by protein- and ligand-observe in-cell NMR approaches. He is also developing novel labeling schemes for human cells, based on the incorporation of fluorinated amino acids and the use of labelled amino acid precursors.
Combined together, these tools provide unique structural and mechanistic insights in a highly physiological cellular setting.
