Lucio Luzzatto -INTERESSI
Lucio Luzzatto ha rivolto continuamente la ricerca e l’insegnamento alla medicina molecolare: vale a dire, a comprendere le malattie umane a livello molecolare, particolarmente nell’ambito delle malattie ematologiche: con il fine ultimo di migliorare la terapia. • LL con i suoi collaboratori, negli anni ottanta, ha realizzato il clonaggio del gene della G6PD, ponendo così le fondamenta per la fisiopatologia molecolare e molti studi popolazionistici della enzimopenia G6PD. Da allora ha continuato a studiare la genetica e la biochimica delle varianti della G6PD, le relative implicazioni cliniche, e le correlazioni genotipo-fenotipo. La sua prima pubblicazione sulla G6PD risale al 1961, l’ultima è del 2024.• Nel settore della malaria, LL e collaboratori hanno dimostrato per primi che la protezione contro la malaria grave è prerogativa delle eterozigoti per enzimopenia G6PD. Inoltre, sono stati compiuti studi sui meccanismi attraverso i quali sia questa enzimopenia, sia la eterozigosi per enoglbina S proteggono dalla mortalità malarica. • Nel settore delle leucemie e linfomi il gruppo di LL è stato tra i primi a studiare i riarrangiamenti dei geni delle immunoglobuline e dei recettori delle cellule T nella diagnosi e nel monitoraggio dei pazienti con queste malattie.• LL ha studiato il tasso delle mutazioni somatiche nella specie umana ed il loro ruolo in malattie.• LL ha svolto ricerca sperimentale sul trasferimento genico avente come fine ultimo la terapia genica, ed ha partecipato alla prima terapia genica efficace della talassemia in un modello murino. • LL e collaboratori hanno per primi dimostrato che l’emoglobinuria parossistica notturna (EPN) è una malattia clonale dell’ematopoiesi. LL ha poi introdotto il concetto di selezione clonale condizionale, oggi ampiamente accettata come patogenesi della malattia. Tale concetto è molto pertinente ai rapporti tra EPN ed anemia aplastica, ed alle attuali terapie dell’EPN. La prima pubblicazione di LL sulla EPN risale al 1970, l’ultima è del 2025.

The leitmotiv of Lucio Luzzatto’s research and teaching has been the understanding of human disease at the molecular level; particularly in the area of blood diseases, for the ultimate purpose to improve their management. LL and colleagues cloned the human G6PD gene in the early eighties, thus laying foundations for the molecular pathophysiology and population genetics of G6PD and G6PD deficiency. In the area of malaria, beyond pioneering the work that indicated that heterozygotes for G6PD deficiency are protected against mortality for P falciparum, LL has opened insights on the mechanism of malaria selection with respect to both the sickle cell trait and G6PD deficiency. LL first provided evidence that paroxysmal nocturnal haemoglobinuria (PNH) is a clonal disorder, introduced the concept of conditional clonal selection and provided supporting evidence: thus rationalising the relationship of clonal expansion with bone marrow failure, and with the treatment of PNH. In summary, LL has devoted his research to contemporary molecular medicine.
